The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer of the IgG fraction is standardized by equivalence-point precipitation. F(ab')2 fragments are tested for purity and specificity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is goat F(ab')2; no trace of albumin, whole molecule IgG or Fc is detected.
Preparation Method
The F(ab')₂ fragment is prepared from the specific goat IgG fraction by limited digestion with pepsin, and purified by gel filtration or affinity chromatography. The resulting F(ab')₂ fragment is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, adjusted to standard titer, filtered through a 0.22 µm filter, vialed and lyophilized. No preservative has been added.
Applications
Product is use as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. It can be used for conjugation with enzymes, radiolabels, fluorochromes or other markers, and for attachment to solid supports for use as an immunoadsorbant. This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. It is also used in Quantitative Precipitin Assay at concentration of 3.0-4.0 mg/mL & in Indirect ELISA(1:80,000).
